XF24 Consumables

The Seahorse XF24 Extracellular Flux Analyzer utilizes three types of consumables including:

FluxPaks simplify the ordering process providing the correct ratio of XF Assay Kits, XF Cell Culture Plates and XF Calibrant to perform up to 18 XF 24-well assays.

XF24 Assay Kit

(available only in FluxPaks)

Assay KitThe XF Assay Kit consists of a disposable, 24-well, dual-analyte sensor cartridge, a calibration plate, and a lid. The measured analytes are OCR (oxygen consumption rate) and ECAR (extracellular acidification rate). Two fluorophores for analyte detection are embedded in a polymer spotted on the end of each of the 24 sensor sleeves. The OCR is determined by the amount that oxygen quenches the fluorescence of a fluorescine complex. The ECAR sensor responds to changes in proton (H+) concentration as they diffuse across the fluorophore based polymetric substrate. The negative log of proton concentration is equal to the pH. The XF Analyzer’s fiber optic wave guides is inserted into the sleeves of the sensor cartridge. Light is pumped through the fiber optic bundles, exciting the fluorophores and the resultant emission is detected and measured. The sensor cartridge contains barcode information matching it to its corresponding tissue culture plate. The disposable assay kit may be used for hours or days. Sensor cartridges are not able to analyze non matching cell culture plates.

Each sensor cartridge also contains 96 drug storage and delivery wells. A unique multi-channel drug delivery system injects 25 to 100µL of up to four different drug compounds into each cell well on command. This allows multiple measurements to be made using a single population of cells, and increases the effective throughput exceeding that of a 96 well plate. XF assay kits are supplied in individual containers and are packaged six per box. XF Assay Kits may only be purchased as part of a FluxPak.

XF24 V7 Cell Culture Microplate

Cell CultureSeahorse XF24 cell culture microplates are designed to maximize the sensitivity of the sensor cartridge measurement. The well design incorporates an initial larger opening with sloping walls that terminate into a well bottom having the same diameter as a 96 well microplate.

Three 200µm high molded stops on the bottom of the plate prevent the sensor cartridge sleeves from contacting or disrupting the cell monolayer. Cells are not affected by the measurement process and can be reused.

A unique feature of the Seahorse XF24 microplate is, unlike other 24-well plates, they are designed to work with commercially available 96-well liquid handling systems.

XF24 cell culture microplates are tissue culture treated for optimum adherence and growth of mammalian cells. The 24-well plates are supplied in individual gamma irradiated containers, and are packaged ten per box. They can also be purchased as part of a FluxPak.

XF24 V28 Cell Culture Microplate

Seahorse XF24 V28 cell culture microplates are designed to increase available oxygen for highly aerobic cells by providing a larger measurement microchamber. The increased microchamber size accommodates islets and tissue sections, too. The larger microchamber increases the dynamic range of the OCR measurement for these cell types, but the ECAR sensitivity is dampened. The well design incorporates an initial larger opening with sloping walls that terminate into a well bottom having the same diameter as a 96 well microplate.

Three 800µm high molded stops on the bottom of the plate prevent the sensor cartridge sleeves from contacting or disrupting the cell monolayer. Cells are not affected by the measurement process and can be reused.

XF24 V7 PET Cell Culture Plate

(Part #101037-004, order separately)

PET plates are identical to the standard V7 plate except the PET plates are molded in Polyethylene Terephthalat, a plastic that does not absorb CO2. PET plates equilibrate faster as a result of less CO2 outgassing and have improved ECAR and CO2 measurements in the XF24 Analyzer.

XF24 Calibrant Solution

Seahorse XF24 calibrant solution is optimized for XF sensor cartridge calibration. The calibrant is pre-mixed and ready to use. A 500ml bottle will calibrate 18 sensor cartridges. The calibrant should be stored at 4°C. The calibrant solution can be purchased individually or as part of a FluxPak.

XF Assay Media

Available August 2008

Seahorse XF Assay Media is specially formulated for XF assays. Just add the required amount of glucose and adjust the pH. This ready-made, unbuffered DMEM formulation saves hours of your time. Each Seahorse certified bottle is supplied with the buffer capacity information which will improve your data quality, while lot consistency decreases variability. XF Assay media is supplied as sterile in 1 liter bottles and packaged two bottles per box.
